血管性血友病因子(von Willebrand Factor, vWF),是由第12号染色体的短臂所编码的糖蛋白。vWF可同时与胶原纤维和血小板结合，当血管破裂时大量血小板以vWF为中介，粘附在胶原纤维上，形成血栓，得以止血。在正常的生理条件下,绝大部份由内皮细胞合成,其余则由巨噬细胞生成。前者产生后多储存在W-P小体（Weibel-Palade body，WPB）内, 后者则储存在血小板的颗粒中。它与一系列心血管疾病如动脉粥样硬化、急性冠状动脉综合征、心房颤动等均关系密切，vWF是一种重要的血浆成分。vWF在止血过程中主要由两种作用：①与血小板膜GPⅠb-Ⅸ复合物及内皮下胶原结合，介导血小板在血管损伤部位的黏附；②与因子Ⅷ结合，作为载体具有稳定因子Ⅷ的作用。此外，vWF也能结合GPⅡb-Ⅲa，参与血小板的聚集过程。正常人血浆vWF浓度约为10mg/L。在内皮细胞受刺激或损伤以及集体处于应激状态时，血浆vWF水平升高。vWF及某些止血与纤溶成分都是血栓性疾病的独立危险因素。相反，当vWF基因发生缺失、插入点突变、剪切点替换或提前形成转录终止信号时，导致血浆vWF量有显著减低或有质的缺陷，不能完成其正常的止血功能时，即为vWD。
