Exonuclease III (E. coli) |

Double-stranded DNA specific exonuclease
Initiates at the 3′ termini of linear double-stranded DNA with 5′ overhangs or blunt ends and 3′ overhangs containing less than four bases
Initiates at nicked sites in double-stranded DNA
Catalyzes the removal of nucleotides from linear or nicked double-stranded DNA in the 3′ to 5′ direction

Exonuclease III is ideal for:

Site-directed mutagenesis
Preparation of single-stranded DNA for dideoxy sequencing
Preparation of nested deletions in double-stranded DNA

Catalyzes the removal of nucleotides from linear single-stranded DNA in the 5′ to 3′ direction
Preferred substrate is double-stranded DNA with 5′ single stranded overhangs > 6 nucleotides long

RecJ_f can be used for:

Degradation of single-stranded DNA from the 5′-end
Removal of 5′ protruding single-stranded termini at the ends of linear double-stranded DNA (blunt ends are not exclusively created)

Removal of incomplete ligation products from ligated circular double-stranded DNA
Degradation of denatured DNA from alkaline-based plasmid purification methods for improved DNA cloning

产品信息

The single-stranded viral DNA is isolated from M13mp18. M13mp18 is a M13 lac phage vector which contains single HindIII, SphI, SbfI, PstI, SalI (AccI/ HincII), XbaI, BamHI, SmaI (XmaI), KpnI (Acc65I), SacI and EcoRI sites within the gene encoding β-Galactosidase.This DNA preparation is useful as a standard and has been tested as a template in the dideoxy-nucleotide termination method of sequencing DNA.

来源

M13mp18 phage is propagated in E. coli ER2738.The phage is purified by polyethylene glycol precipitation. The single-strandedDNA is then extracted with phenol.
