病毒灭活和稳定缓冲液（VIB）Inactivir®PLUS（符合REACH标准）
 lifescienceproduction病毒灭活和稳定缓冲器InActuix®Plus（REACH兼容）已经与贝福德郡大学合作开发。2019冠状病毒疾病是一种胍基和基于Destin的失活缓冲液，基于已公布的数据，已形成缓冲，该病毒将灭活病毒，如COVID19和季节性流感和流行病，如SARS、MeS和埃博拉。


本产品用于在含有患者或其他潜在致病性样本的输送管中灭活病原体。它会分解细胞，使样本中的任何病毒变性，使病毒失去活性和非致病性。这就产生了用于运输和测试的非致病性样本，另外一个优点是，由于可能存在的降解酶变性，病毒样本是稳定的。样品不需要保持在+4°C，从而消除了搬运过程中对冷链的要求，从而大大降低了运输成本。


使用VIB Inactivir®PLUS（REACH兼容）中采集的样本将通过降低潜在致病性样本的2+/3类处理要求，提高检测速度和易用性，从而提高实验室吞吐量。该VIB与标准的基于二氧化硅的RNA提取协议广泛兼容，并且可以在不中断当前处理协议的情况下使用。经VIB Inactivir®PLUS（REACH兼容）处理的样品也可以直接用于基于二氧化硅的RNA提取方案，而无需在第一个提取步骤中添加进一步的变性缓冲液。这样就可以将样品直接添加到粘合材料中，而无需稀释。由于纯化的样本量增加，这简化了RNA提取协议并提高了灵敏度。


病毒灭活缓冲液Inactivir®PLUS（REACH兼容）已开发出一个优化的缓冲系统，以提高RNA稳定性。为确保REACH合规性，使用了Triton X100的替代洗涤剂，并经过验证，以确保功效不会受到影响。

概述

T-STORE®（组织储存）是一种无菌、化学成分明确且经过优化的培养基，开发用于为哺乳动物组织样本和细胞提供稳定的储存和运输环境。该产品设计用于维持组织样本的完整性，从而减少坏死和细胞凋亡。

  • TORE®采用稳健的缓冲系统，允许在较宽的温度范围（+ 2 ℃至

+ 37 ℃）。此外，它被设计为与人血清和间质液等渗、等渗和等离子，根据需要创造一个适合维持离体组织和细胞数小时或数天的稳态环境。

 

使用T-STORE®的优势

  • T-STORE®是一种“即用型”运输和储存介质，适用于采集各种细胞和组织。
  • T-STORE®为高度时间关键工艺提供了灵活性。
  • T-STORE®可在亚常温（非冷冻）和/或常温温度下使用。

应用程序

分析前组织样本的储存和运输

进一步分析前，分离/分离或富集后细胞的储存组织

T-STORE®模拟间质液的基本组成，意味着在储存和运输过程中维持组织的酸碱(pH)平衡，允许组织在清除后72小时内成功使用。在T-STORE®中储存可使组织样本保留遗传和组织学特征长达72小时。

虽然其他组织保存液具有“组织特异性”，但T-STORE®可用于各种类型的正常和恶性组织，是储存和运输“新鲜”样本进行进一步处理的理想选择。

细胞支架、储存和运输

T-STORE®提供了理想的液体成分，可保持离体人体细胞的酸碱平衡和完整性。该制剂的*之处促进了三磷酸腺苷(ATP)生成中的线粒体代谢。

T-STORE®适用于无数干细胞和外周血细胞类型，无需添加破坏性化合物，可‘冷’（低温）、亚常温（+ 15 ℃～

+ 25 ℃）和常温条件下。该产品也有可能用于细胞培养生物反应器。

使用T-STORE®

组织样本的获取

T-STORE®以纯品形式提供，无需进一步稀释。其他抗生素制剂可用，包括两性霉素B和氯霉素以及两性霉素B和纳米霉素。作为生产商，我们还能够根据客户质量标准为T-STORE®提供定制添加剂。产品介绍请参见下表2。

切除后，组织样本应立即*浸没在T-STORE中。所需的T-STORE®体积应由用户确定，如样本量和

 

下游应用程序可能会影响需求。作为进一步优化的起点，我们建议使用无顶空的无菌防漏容器*浸没在T-STORE®中。建议使用过量的T-STORE®，以确保成功储存。

使用T-STORE®在室温下储存细胞

储存前，应在推荐条件下培养细胞系。

  1. 分离生长活跃的贴壁细胞或直接收获悬浮细胞，计算细胞数量和活力。
  2. 室温下以180 xg沉淀细胞5 min，弃去上清液。将沉淀物重悬于3 mL T-STORE®中，并按照上文所述进行重悬。弃去上清液。
  3. 将细胞团块重悬于2 mL T-STORE中，并转移至2 mL冷冻管（或等同产品）中。
  4. 试管可在室温下避光储存长达72h。稳健的细胞类型可储存长达96h。
  5. 为回收细胞，在转移至含有*生长培养基的适当容器中之前，应轻轻重悬细胞（回收前无需移除T-STORE®）。在最佳条件下（例如，在37 ℃、5%CO2的加湿培养箱中培养细胞）。
  6. 应在48h内观察到细胞*恢复。 

 

作为质量控制过程的一部分，每批T-STORE®在放行前均进行了功能性检测。功能测试使用HEK细胞和上述方案。显示的数据已从14个批次中整理，并显示在T-STORE中储存72h后，细胞数量保持在好活力和回收率。

 

 

规格

 

室温下在工作台上放置96小时后，储存在T-STORE®（C和C）中的HEK细胞保持粘附，并在储存前保持与细胞相似的形态（上图）。储存在DMEM/HEPES中的细胞形态发生改变，96小时后失去粘附性（B和B）。对照细胞（A和A）在37 ℃、5%CO2的*DMEM中培养，在96小时内过度生长。图像来自两个独立的实验（大写和小写标签）。原始放大率100X。

 

T-STORE®是一种无菌即用型溶液。T-STORE®可与抗生素联合使用：两性霉素B和氯霉素或两性霉素B和纳米霉素。
