产品介绍 磷脂酰丝氨酸(PS)是一种带负电荷的磷脂,正常细胞中,PS只分布在细胞膜脂质双层的内侧,而在细胞凋亡早期,细胞膜 PS由脂膜内侧翻向细胞膜外侧,使PS暴露在细胞膜外表面。Annexin V是一种分子量为35~36kD 的Ca2+依赖性磷脂结合蛋白,与磷脂酰丝氨酸有高度亲和力,故可通过细胞外侧暴露的磷脂酰丝氨酸与凋亡早期细胞的胞膜结合。因此Annexin V被公认为检测细胞早期凋亡的灵敏指标之一。
将Annexin V进行FITC标记,以标记了的Annexin V作为探针,利用荧光显微镜或流式细胞仪可检测细胞凋亡的发生。碘化丙啶(Propidium Iodide, PI)是一种核酸染料,它不能透过正常细胞或早期凋亡细胞的完整的细胞膜,但对凋亡中晚期的细胞和坏死细胞,PI能够透过细胞膜而使细胞核染红。因此采用Annexin V与PI双染的方法,就可以将处于不同凋亡时期的细胞区分开来。

Jurkat cells (T-cell leukemia,human) treated with 10 μM of camptothecin for 4 hours(panel right) or untreated control(panel left).
1. Wash cells twice with cold PBS and then resuspend cells in 1×Binding Buffer at a concentration of 1×106 cells/ml.
2. Transfer 100 μl of the solution (1×105cells) to a 5 ml culture tube.
3. Add 5 μl of Annexin V-FITC.
4. Add 5 μl PI.
5. Gently vortex the cells and incubate for 15 min at RT (25°C) in the dark.
6. Add 400 μl of 1×Binding Buffer to each tube.Analyze by flow cytometry within 1 hr.

Annexin V is a calcium-dependent phospholipid-binding protein that has a high affinity for PS, and is useful for identifying apoptotic cells with exposed PS. Propidium Iodide (PI) is a standard flow cytometric viability probe and is used to distinguish viable from nonviable cells. Viable cells with intact membranes exclude PI, whereas the membranes of dead and damaged cells are permeable to PI. Cells that stain positive for FITC Annexin V and negative for PI are undergoing apoptosis. Cells that stain positive for both FITC Annexin V and PI are either in the end stage of apoptosis, are undergoing necrosis, or are already dead.

产品介绍 免疫荧光标记技术(immunofluorescence technique)是将已知的抗体或抗原分子标记上荧光素,当与其相对应的抗原或抗体起反应时,在形成的复合物上就带有一定量的荧光素,在荧光显微镜下就可以看见发出荧光的抗原抗体结合部位,检测出抗原或抗体。该技术的主要特点是:特异性强、敏感性高、速度快。
荧光素标记抗体简称荧光抗体(FA),是目前广泛用于免疫病理、细胞化学、流式细胞学、病毒学及自身抗体的临床免疫诊断之中的特异、灵敏、定性和定位相结合的免疫化学试剂。用于标记的抗体,要求是高特异性和高亲和力的。所用抗血清中不应含有针对标本中正常组织的抗体。一般需经纯化提取IgG、IgM后再作标记。
本试剂盒中的荧光素采用高质量进口的异硫氰酸荧光素(FITC),FITC标记抗体的原理是利用FITC上的N=C=S基团和抗体上游离的-NH2发生化学形成FITC-抗体结合物,即荧光抗体。一个IgG分子中有86个赖氨酸残基,一般最多能结合15-20个,一个IgG分子可结合2-8个分子的FITC。

产品介绍 Fluorescein isothiocyanate (FITC) is a derivative of fluorescein used in wide-ranging applications including flow cytometry. FITC is the original fluorescein molecule functionalized with an isothiocyanate reactive group (-N=C=S), replacing a hydrogen atom on the bottom ring of the structure. This derivative is reactive towards nucleophiles including amine and sulfhydryl groups on proteins.
A succinimidyl-ester functional group attached to the fluorescein core, creating “NHS-fluorescein”, forms another common amine reactive derivative that has much greater specificity toward primary amines in the presence of other nucleophiles.
FITC has excitation and emission spectrum peak wavelengths of approximately 495 nm/519 nm, giving it a green color. Like most fluorochromes, it is prone to photobleaching. Because of the problem with photobleaching, derivatives of fluorescein such as Alexa 488 and DyLight 488 have been tailored for various chemical and biological applications where greater photostability, higher fluorescence intensity, or different attachment groups are needed.

鬼笔环肽(Phalloidin)是一种来源于毒蕈类鬼笔鹅膏(Amanita phalloides)的环状七肽毒素,以高亲和力(Kd= 20 nM)选择性结合于丝状肌动蛋白F-actin,而不会与单体肌动蛋白G-actin结合,通常用来标记组织切片,细胞培养物或无细胞体系中的F-actin,从而对F-actin进行定性和定量分析。另外,鬼笔环肽衍生物也以相近的亲和力结合于大小纤维,无论是动植物来源的肌肉细胞或非肌肉细胞,按照每一个肌动蛋白亚基约与一个鬼笔环肽分子的计量比结合。且非特异性结合几乎可忽略,染色区域和非染色区域辨识度非常明显。因此,鬼笔环肽衍生物特别适合替代肌动蛋白(Actin)抗体进行相关研究。另外鬼笔环肽衍生物很小,直径约12-15Å,分子量<2000 Daltons,未标记肌动蛋白(Actin)的许多生理特性都得以维持,比如,同肌动蛋白结合蛋白如肌球蛋白,原肌球蛋白,DNase I等仍能发生反应;鬼笔环肽标记的纤维丝仍可穿透固相肌球蛋白基质;以及甘油抽提的肌纤维标记后仍可收缩等。
鬼笔环肽(Phalloidin)的结合阻止丝状肌动蛋白(微丝)的解离,稳定微丝结构,从而破坏微丝的聚合-去聚合的动态平衡。此特性使得肌动蛋白聚合发生的临界浓度(CC)降至<1µg/mL,因此,可用作一种聚合促进剂。此外,鬼笔环肽还可抑制F-actin的ATP水解活性。
本品为FITC标记的鬼笔环肽,染色反应特异性强,对比性高,具有比Actin抗体更好的染色效果,适合用作F-actin的定性和定量检测。另外,经本品结合后的F-actin仍能维持actin自身具有的许多生物学特性。且本品的结合没有物种差异性,适用性广泛。

工作液配制:
本品为 20µM 储存液,总量为 300µL。按照 100 nM 的工作液浓度来换算,可制备总量为 60 mL 的工作液。建议收到产品后,根据单次使用量分装保存,-20℃避光冻存,一年稳定。
实验前,用 1×PBS稀释储存液到需要的工作浓度。推荐工作浓度为:80~200nM(1:100-250 倍稀释),工作液现配现用。最佳稀释比例可以根据实际染色效果进行适当调整。

染色步骤:
1. 细胞爬片生长 24h,使其密度达到 50%汇合度。
2. 吸掉培养液,37℃预热的 1×PBS(pH 7.4)清洗细胞 2 次。
3. 使用4%甲醛PBS溶液进行细胞固定,室温固定 10min。
注:避免固定剂中含有甲醇成分,因为甲醇在固定过程中可能破坏肌动蛋白。
4. 室温条件下,用 PBS 清洗细胞 2~3 次,每次 10min。
5. 室温条件下,用丙酮(≤-20℃)脱水或者用 0.5% Triton X-100 溶液透化处理 5min。
6. 室温条件下,用 PBS 清洗细胞 2~3 次,每次 10min。
7. 取 200µl 配制好的 FITC 标记鬼笔环肽工作液,覆盖住盖玻片上的细胞,室温避光孵育 30min(通常情况下,4℃~37℃ 孵育皆可)。
注:为了降低背景,可于 FITC 标记的鬼笔环肽工作液内加入 1% BSA;另外,孵育过程中为了避免溶液挥发,可将盖玻片转移到一个密封的容器内。
8. 用 PBS 清洗盖玻片 3 次,每次 5min。
9. 用 200µl DAPI 溶液(浓度:100 nM)对细胞核进行复染,约 30s。
10. 用 PBS 清洗盖玻片,然后倒置在已经滴有一滴 Fluoromount-GTM 水溶性封片剂的载玻片上。使用纸巾轻轻檫掉多余封片剂,然后用指甲油永久封片。此法制备的标本玻片可置于 4℃避光保存,通常 6 个月内可继续做 F-actin 染色分析。
注:也可以直接使用含有 DAPI 的抗荧光淬灭封片剂合并步骤 9. 10。
11. 荧光显微镜或者共聚焦显微镜下进行荧光观察,FITC(Ex/Em=496/516nm),DAPI(Ex/Em=364/454nm)。

注意事项:
1. 鬼笔环肽具有毒性,需小心操作(对人的半数致死剂量 LD50 约 2mg/kg)。
