优势

·¨Cypridina荧光素酶和Gaussia荧光素酶在转染细胞的上清液和裂解液中都具有非常强的信号。因此,可作为双重分泌报告者或双重细胞内报告者使用。

·¨天然鲤鱼(Vargula)荧光素酶和高斯荧光素酶具有天然分泌信号,表达后有效分泌到细胞培养基中。测定荧光素酶不需要细胞裂解。

·¨Cypridina荧光素酶比萤火虫/肾素荧光素酶亮20-30倍,具有最高的转换数和比常用萤火虫荧光素酶更高的生物发光信号强度,使其成为理想的转录报告者(1)。

·¨Gaussia荧光素酶分析系统的稳定剂组件在较长时间内提供稳定的动力学,为用户提供高通量分析和手动交付分析所需的时间。

·¨分泌的塞浦路斯荧光素酶蛋白稳定,具有*的光产生活性,允许进行非常敏感的分析(1,2)。

·¨同时含有CLuc和分泌型高斯荧光素酶(即转染后的生长培养基或细胞裂解物)的样品可在-20°C下长期储存。

描述:一组经过改进的超敏感分泌型荧光素酶报告子已被开发出来,用于分析同一组转染细胞中不同的启动子活性。这种方法不仅可以分析同一组细胞中的三条或多条通路(反应),而且还可以在不杀死细胞的情况下实时研究反应(因为这三条反应器是分泌的)。靶向系统提供了许多不同的细胞内和分泌型双荧光素酶分析试剂。Cycprida–Gaussia荧光素酶分析系统特别有吸引力,因为Cypprida荧光素酶和Gaussia荧光素酶在细胞内和分泌部分都具有强大的活性。当使用该试剂进行分析时,它们都具有稳定的生物发光信号。

塞浦路斯(瓦古拉)荧光素酶:

Cypridina荧光素酶(以前称为Vargula荧光素酶)来自海洋介形类Vargula Hilgendorfi或Cypridina Noctilucus,是一种分泌型荧光素酶,最大发射波长为460nm。它的亮度大约是萤火虫荧光素酶的20-30倍。拥有最高的营业额。与萤火虫荧光素酶不同,Cypridina-lcufierase不需要ATP,并催化其*底物Cypridina-luciferin的氧化,如前所述

Cypridina荧光素不同于腔烯特拉嗪,腔烯特拉嗪是雷尼拉、高斯和Metridia荧光素的底物。由于其*的底物和明亮的分泌型荧光素酶活性,Cypridina荧光素酶在涉及Gaussia、Renilla或Firefly荧光素酶的多重检测中特别有用。分泌型CLuc是一种非常稳定的蛋白质。由于这一特性,从上清液中测得的活性反映了在取样之前累积的蛋白质量。塞浦路斯荧光素酶是由细胞自然分泌的(图2)。因此,细胞裂解对于荧光素酶活性的测定是不必要的。

高斯荧光素酶:关于高斯荧光素酶:

高斯荧光素酶(GLuc)是海洋桡足类高斯荧光素酶(1,2)。这种不需要ATP的荧光素酶在产生光的反应中催化底物柯伦他嗪的氧化,与其他发光报告基因相比,它具有相当大的优势,例如分泌性和更亮的信号强度,以及生物发光信号的稳定性(约10%在一小时内衰减)。从转染了表达GLuc的质粒的培养细胞上清液中测得的发光与产生的酶量成正比,这反过来又反映了转录水平。或者,可以使用细胞裂解物进行分析。虽然大部分活性是分泌的,但高斯荧光素酶的高灵敏度也允许从细胞裂解物中进行测量。

该试剂适用于需要分析大量样品的高温超导应用。在没有稳定器的情况下,信号强度最初略高,但衰减速度比有稳定器时快。
注:数据是在Turner TD2020光度计上测量的三次测定的平均值。

We began in 1948 as Normal Wood Products, a small, regional supplier in Midland, Michigan, and have grown to become a nationally recognized caseworks producer and market leader.

In the mid-1990s, Bob Bowden and Dave Clark bought the company and began to build the Case Systems brand. Bowden and Clark expanded our company's sales efforts and provided additional capital, all of which accelerated our rapid growth. We soon outgrew our facility and moved to our current location.

Case Systems added BOSTONtec to the Case Systems’ family in 2002. BOSTONtec offers a full complementary of flexible table systems and overhead service distribution solutions that define the space for Research & Development laboratory space.

In November 2011, the portfolio was expanded to include this premium wood casework manufacturer, CiF Lab Solutions. CiF provides “Best-In-Class” wood casework that is designed, produced and installed to complete an AWI Premium installation offered in several selections of wood veneer species.

Today Case Systems continues to grow and expand our business model to meet and exceed the needs of our customers.
