Athrogen-CIA®致关节炎单克隆抗体合剂——胶原抗体诱导关节炎（CAIA）法必选

早在1998年，人们就在应用胶原的抗体来诱导风湿性关节炎（Collagen Antibody-Induced Arthritis (CAIA)），chondrex公司早期的产品主要是四个抗混合试剂盒。为了减小模型之间的差异，并增加不同品系小鼠的诱导效果（包括CAIA低反应性的C57BL/6小鼠、C57BL/6背景的基因敲出小鼠和转基因小鼠）。Chondrex公司开发出5种单克隆抗体混合的试剂盒，为更好的诱导风湿性关节炎模型提供了有力的支撑。

二、应用

用于小鼠关节炎诱导（CAIA），此模型是研究类风湿性关节炎发病机制和治疗药物筛选的理想模型。

三、产品信息

（1）Kit由小鼠mAb合剂和LPS组成：

mAb合剂是抗II型胶原蛋白的5个单克隆抗体的混合物，包括clone A2-10 (IgG2a)、F10-21 (IgG2a)、D8-6 (IgG2a)、 D1- 2G(IgG2b)、D2-112 (IgG2b)，识别多种物种中Ⅱ型胶原蛋白的保守抗原表位。其中，Clones A2-10、D1-2G、和D2-112识别在II型胶原蛋白CB11片段(124-402)上的167个氨基酸片段LyC1 (124-290)簇集的抗原表位，Clones F10-21和D8-6识别II型胶原蛋白的CB11片段 (124-402)上的83个氨基酸多肽片段LyC2 (291-374)上簇集的抗原表位。

（2）mAb来源于小鼠，溶解于PBS中，pH 7.2，浓度为10mg/ml。

（3）LPS是内毒素的一种，来自E. Coli 0111:B4，用于刺激免疫反应，生成动物模型，如果不用LPS免疫，关节炎的严重程度会降低。

四、适用动物

7-8周龄或更大的小鼠

（1）CAIA高敏感性小鼠：DBA/1、Balb/c、B10.RIII、 C.B-17、scid/scid或129/Sv

（2）CAIA低敏感性小鼠：C57BL/6或C57BL/6背景基因构建小鼠

五、交叉反应性

与大多数种类的Ⅱ型胶原蛋白可以交叉反应，包括：人、大鼠、小鼠、鸡、兔子、狗、猪、牛、马、猴子（D2-112不和鸡Ⅱ型胶原蛋白反应）。

六、用法

（1）联合使用单克隆抗体混合物和LPS来诱导CAIA敏感性小鼠关节炎（(DBA/1、Balb/c、B10.RIII、C.B-17、scid/scid或129/Sv)）

第0天：静脉或腹腔注射1.5mg的5-克隆混合物。

第3天：腹腔注射25-50ug的LPS 。

注意：在3-4天时可以观察到关节炎由温和至严重，7-10天时达到最严重。

（2）联合使用单克隆抗体混合物和LPS来诱导低反应性小鼠关节炎（C57BL/6 或 C57BL/6背景基因构建的小鼠）

第0天：静脉或腹腔注射5mg的5-克隆混合物。

第3天：腹腔注射25-50ug的LPS。

注意：在3-4天时可以观察到关节炎由温和至严重，7-10天时达到最严重；可在10-14天时再次注射LPS再次刺激炎症关节炎。

（3）利用单克隆抗体混合物诱导DBA/1 和Balb/c小鼠关节炎

第0天：静脉注射6-10mg的5-克隆单克隆抗体。

注意：在2-3天时可观察到关节炎。此种方法在其他的CAIA敏感性小鼠中还没有得到验证

小鼠对单抗合剂和LPS的易感性受品系、饲养条件、个体差异及动物的处理和生存环境等影响。建议SPF级条件，因为传统条件时，细菌污染可能使动物产生免疫反应，只产生轻微的关节炎。建议在大规模实验前，先用小样本量做预研究。研究者应根据自身实验需求，选用最合适的免疫程序。
